How we can know whether we have connected to the
Sapdb instance or not?

What do you mean? The sapdb instance of you portal?
What OS are you using? If Windows you should be able to see that in your Microsoft Management Console. The java server instance won't be able to start if it's not connected to the database. You could also use command line tools (e g dbmcli) that you get when you install your sapdb instance.
