1.) When trying to run SXMB_IFR the followng message is coming.
2.) When trying to run the transaction SPROXY I am getting a mesage " No connection with Integration Builder."

Regarding SPROXY: For Proxy scenarios you need to perform basic steps in application server(R/3)
Steps: Needd to set up a HTTP connection with XI and thus you will be able to see IR objects for Proxy generation.

For you first Ques, if you are not able to open the Integration Builder page by using SXMB_IFR, then
1. Check your firewall settings.
2. Alos check if the user have corresponding roles (using tcode SU01).

sure you need to perform Post Installation steps plus need to make your Application Server ready for Proxies..

Which version of java have been installed on your machine?? If it is hiegher then it will create problems. You should use j2sdk_1.4.2_10.
Furthermore ensure that the required JDK is being used in the client system.
Set the proxies to NONE in the Java Web Start for this use the below path
Start>Programs>Java Web Start>File>Preferences-->Proxies

Check your RFC destinatoin using the Transaction sm59 and see if your integration server is properly configured in the sources/destination system..
