Go to SM59 , TCP/Ip Connections:
Check OPTSERVER_ Entries...These need to pass the connection test.
Once these would pass the connection test optimizer would be up.. Make sure the settings of these connections points to correct .exe file in Installation Directory of your Optimizer.
Contact your BASIS to get the path for these files.

There are little problems with the TVSR01 engine (tr.RCC_CUST). The fact is that the RFC connection OPTSERVER_TVSR01 has incorrect parameters in technical settings (tr.SM59). You need to check the target host of your SAP TM and the path of vsropsvr.exe . The program vsropsvr.exe must be have the correct path for your SAP TM system. Your basic support team can to clarify this path. Check the every engine.
And so one remark for the Load Optimization engine. Initially set the wrong engine. You must specify the program 'vsoopsvr.exe' to the RFC 'OPTSERVER_TVSO01'.
