Is it possible default the storage unit type in LT04 ?

if yes, kindly advise how to configure in SPRO?

Transaction LT04 is to create a Transfer order from a Transfer requirement

There is no specific SPRO setting related to LT04 you need to explain why you need that probably based on that we can suggest alternate, if you want to overwrite data of TR then there is no other standard approach other then writing your own piece of code in user exit. Also as mentioned by Jurgen as per standard at the time of TO creation system check the palletization data (WM2) view from material master and based on that it propose the SUT

Not sure if you use HU and packing material. You can try through this config

IMG-Logistics General - Handling Unit management - Basics - Warehouse Management  - Assign Storage Unit type to Packing Material Type.

For every packing material you can maintain a default storage unit type
