if the reason for (re)configuring TMS is a system copy, check if secure storage is consistent(TA SECSTORE). If the store is corrupt, system will not be able to execute RFC's, not even to itself.

Issue DICO tp delete TMS configuration.
Make sure permissions, UID, GID and other things are mapped as per the guide and documents.
Logon to your domain controller and reconfigure TMS.

I really accept what RajSekhar Reddy suggested but on your scenario ,you have to create the
RFC and STMS configuration from the 000 Client in the Application Server once again and ,please be sure checking before deleting the RFC connection and restart the Instance once ,i think that your RFC and STMS are not configured they are in default condition
If they are configured attach the TMS route DEV--->QAS--->PRD Screenshot

For this U have to delete Your TMS configuration, and u have to create it again .
TMS once again by these steps

It seems the Problem with RFC connection...
Please check Authorization test for the the respective RFC connection. Utilities --> test --> Authorization test.
if fails the connection test, provide user & password in the logon & secutiry Tab of connecton.

Error relates with permissions on trans directory.Please check permissions on the complete E:\usr\sap\trans directory and retest with RSTPTEST.
