Hi ,
We are using SAPECC6 EHP3 with Mssql2005 and windows 2008r2 OS.
DEV and QAS servers are virtual servers.
PRD server with Windows cluster (we have 2 servers sapsrv01,sapsrv02(cluster drives are in sapsrv02).
We are planing to upgrade my database to mssql2008r2 (for database compression)
i want to know below information.
1)How to upgrade my database with any affect to SAP.
2)How to do the Database upgrade in windows cluster environment.
If any one did this activity please share me the documents.
Please provide the detailed information.

I believe, that here you mean not to make any changes such as change in release or SP level.
For this check PAM for product support and relevant notes

1144459 - SAP Installation Media and SQL4SAP for SQL Server 2008 (R2)
2)How to do the Database upgrade in windows cluster environment.
Refer:

And Update ODBC drivers
You may have to update your kernel with DBSL as well.

Database upgrade has to be performed with application down to avoid any impact.
Secondly read relevant upgrade document available at SAP service marketplace to check out for pre-requisites before the upgrade.
You can use SAP tools for MSSQL to perform the required upgrade
