Is there any way we can upgrade all the instances at once.

also SUM have a option to select 'upgrade dialog instances' . more details in SUM guide
You don't upgrade application servers. The upgrade is done on the primary application server (Previously Central Instance).
If this is not the answer you are looking for then you may provide more details about your situation.

Thanks Reagan, In our case we have around 70 XMII SAP systems, I was wondering is there any way we can upgrade all these systems.

If the product is "SAP xApp Manufacturing Integration and Intelligence" then I don't think SUM is the upgrade tool used here as the upgrade guide doesn't speak about SUM usage.
