1. In STMS before selecting the system just do the refresh the system landscape.
2. kindly check the SAP Note 98678 - SQL Server Connection Issues
3. Logon to Domain controller just do the refresh & update the configuration once

Check whether the target system kernel files are on the same level.
Also check whether this error is happening for all the transports or only for this transport request.

Please test the transport connection using STMS -> Check->Transport tool for the affected system.
I assume you already performed kernel upgrade to patch 225 during which some dbsl libraries are lost.
Could you retain the old kernel verson and overwrite the new kernel version.
Post this check the results.
