Hello -
In our environment we have the domain controller on QA box, that talks to both DEV and PROD, but the PROD does not directly talk to DEV due to firewall rules. Should that be a concern and why?

As long as the transports work, then the set up you have so should not be an issue.

Firstly, the scenario is wrong as production clients should not be communicate to DEV and QAS directly. Because the object move from DEV->QAS to PRD not PRD to DEV.
Secondly PRD clients creation is always commanded as readable(no modification possible).
Third is the TMS configuration time we never define the transport route from PRD to QAS or PRD to DEV.
If you want to take some object from PRD. Then we can take,but we must need to change PRD clients.
Client Approval is required to do the above activity.

It should not be an issue till the time you have SAP's standard connectivity working (RFCs etc.).
I am interested to know why there is a security restriction?

You can disable the firewall and than check the communication between DEV to PROD.
