To solve this issue, try below steps:
1. In the web UI of process tree, change the Activity Display from "Standard" to "Extended".
2. Execute the activity "Define RFC Destination".
3. Choose the button "State" at the bottom of the screen.
4. If there are some entries, select the entries and choose the button "Reset" and then choose "Back".
5. Choose button "Synchronize" in the main Window.
6. Your RFCs should be fine now.

Hello,
Which SP you are using ? Juts try to re-execute the define RFC activity once and if it still does not work, then check for any logs in SM21 and dumps in ST22, and update the same to analyse further.
