I believe the SAP Note 1759273 - Cache refresh fails should fix your issue. I see the exact error message.

Just make sure proper authorization is in place..
Also check the RFC destination 'INTEGRATION_DIRECTORY_HMI' should have path prefix as /dir/CacheRefresh. Do a connection test.
