I have one question. vendor claims that he has not recived the purchase order, but as I see in SCOT, there is no pending communication.
Now how do I ensure that PO is sent to vednor?

I would recommend you to check into SOST transaction code in your system and verify the PO output successfully triggered or not? SCOT does not give that information.
If your SOST shows correct status (i.e. Green) that it send out successfully that means Email from SAP system went to your client Mail server successfully.
Now next step is to verify if your Mail server was working at the time SRM send PO? There may be chances that mail server was temporary down and they did not receive the message. In that case as Govardhan mentioned re-trigger output again, You can do it from SOST when you find the PO email you can re-trigger the same.
If your Mail server communication was up and running then request vendor to verify their Junk or Spam Folder. This happen when Vendor mail server have some spamming filter, In one of the client i have encountered issue where Client Mail domain was put on Black List by one of the Vendor's SPAM checking Anti -Virus. We spend almost entire week figuring out by checking with Vendor IT department and finally got the solution...

Hi,
Have you seen the output status, please check the status of output via SRM PO display under Header details you can find 'output' option where you should be able to see the status as 'success' /not processed or failed and reasons for that.
If it is successful, take the time and search in scot for output info. Also you can re-generate the output once again.
In case, if it is the first time PO for that vendor please check the output settings for that vendor via BBPMAININT in SRM GUI or in Administration through portal.
