Hi Experts,
we have an XYZ interface,we have already configred a path for HCM inbound files.Can the same interface poll another directory also, For ISP?is it technically feasible?

I dont understand your requeriments well, you should to explain your goal more cleary, are they for sender or for receiver?, Respect for receiver your requeriments i find several solutions:
A) You can add to the same service interface several communication channel, in every channel you can put the path that you want.
B) If you want to reuse the same channel but you want that this channel can deteminate different paths taking a value from payload or from soap header you can use variable substitution. C) If you want that one message goes to diferent paths you can use the multimapping in order to split the original message in various and you can comnbine with the option a or the option b. Check this document http://scn.sap.com/docs/DOC-3820
For the sender you can use too patterns to search in several places or to put several paths:
