Once you have established VPN connectivity, the machine becomes part of the network [that is the Virtual Private Network] just like ay other machine.
As long as the VPN is maintained NCo will work as expected.
in this scenario, if you are using a dynamic IP, you will need to write a script that recognizes the IP assigned to the server. then it should publish this IP on the network at a fixed location, and this can then be read by the consumers of the services which will append the remaining part of the url after the ip address
to answer your question simply :
yes, it will work as long as the server is part of the network [VPN is active] and the IP address is known to the consumers of the services hosted
