Lookup's arfe nothing but perfroming Cross Referencing from withing your Message Mapping itself.
Consider a case, when you get a value from a source and you need to use this value to get a corresponding target value from an R3 table. You can use RFC Lookups in these cases to make an RFC call from the message mapping itself. Likewise, you can perfrom DB look up to get data from a database and a SOAP Look up to call a webservice, More on thse blogs,
