My friend is installing NW development studio and working on Duet(earlier Mendecino).He has a doubt -:
1. what is the port through which net weaver communicates with mySAP (especially WAS) ?

There's also a great Network Integration document attached to SAP Note 21151 which has not only the ports, but some nice diagrams and explanations of the overall network architecture & integration points.

there are a huge amount of different ports used by a WebAS.
In developer studio, 2 of them are for interest, the message server port and the j2ee http port.
message server port = 36+instancenumber, e.g. 3600 for instance 00
j2ee http port = 5instancenumber00, e.g. 50100 for instance number 01
if you need other ports, just ask
