PO's are transferred from ECC to SCM through standard proxy. When PO XML message starts to process, per PO Line check if Lane exist.If does not exist, email Support
How and where to check and do the validations.??

There is a validation check "PO_LANE_MISSING" which checks a transportation lane exists in SAP SNC between the ship-from location and the customer location.
This validation check is by default maintained with warning priority.
Path:SPRO>Supply Network Collaboration>Basic Settings>Validation>SAP Standard Settings-->Display Settings for Standard Validation Profiles
Validation profile:POC1
Validation check:PO_LANE_MISSING
Now if your requirement is whenever PO fails in SNC with transportation lane is not maintained then you need activate first alert type 7035 and create email alert notification profile:
1)Activation of alert type 7035:
Path:SPRO>Supply Network Collaboration>Exceptions>Alert Type Activation>Activate Alert Types
Alert type=7035

save this entry.
2)Creation of email alert notification:
See the below link:
In the above link you need to change alert type from 7051 to 7035.
Once above setting is done user will get email with PO#,supplier,plant and product whenever PO gets failed in SNC.
