Hi:
The right thing is to have a device profile assigned to the device with JRE, SETUP_WIN32, DB2E and APP although there is not a setup package? Or with only the application is sufficient for the device profile?
For example if you wanted to install a new version of SP (SETUP_WIN32) could be added directly to a device profile or should be exist previosly?
My installations is in 7.1 SP11, LAPTOP, WIN32.

Setup packages are used only during initial roll out of devices. They cannot be used for upgrade scenarios. Check following link:

Coming to your question, it is not mandatory to create a setup package. You can create device profile and assign all required mobile components and can then assign this device profile to devices. Check link below which also explains about upgrade scenario:
