So far I look at Wily Workstation and drill down to each instance to check.
1) I want to set Wily to monitor some JAVA systems by itself.
2) The monitoring results shold be emailed to the support desk.
Could you please tell me how to configure it?

You could use the System Monitoring tool in the Solution Manager Workcenter to accomplish this task, but I'm not sure you will be satisfied by it since this functionality is more generic than the Workstation.
Based on your description you are already familiar and you already have all the metrics int he Wily Workstation. I'd recommend purchasing the full version of Wily EM you will be able to configure alerts and customize the dashboards. Check note 797147 to contact the person responsible for licensing.

We already have WilyEM. I have access to both Wily Workstation and WebView.
What I need is a quick proof of concept: Once the Java system (Portal)'s response time is longer than
500ms(for example) an email will be sent.
I only did this for ABAP at RZ20 before.
Could you please point to some links with details about how to set it up?

The Wily version delivered by SAP doesn't include as full license.
You cannot customize the alerting without a full license.
All the details are explained in Note 1134542.
Answering to your use case, yes, it is possible.
More details are contained in the Configuration and Administration Guide for CA Wily Introscope.

There are two types of Alerts: Simple and Summary.
-> A Simple Alert takes status information produced by a comparison as input, compares it to user-defined threshold values, and outputs a status.
-> A Summary Alert bases its status on the status of multiple Simple Alerts and other Summary Alerts.
Both types of Alerts appear together under the Alerts node in the Investigator tree.
