how do you distinguish between standard document type NB and newly document type customised ZNB for screen layout.

This can be checked from User Settings. In Easy access screen go to Options>Expert Tab>Controls, Check on Show Keys on All Drop down Lists and apply the settings.
When you will open PO Screen (ME21N/ME22N/ME23N), in the Document Type Field the NB will be showing as Prefix for Standard PO and any other PO Doc Type (if maintained in Customizing) say, NB Standard PO , PO01 Domestic PO , PO02 Import PO etc.

If you know what is available for standard document type NB,You can easily identify & configure screen layout according to things need to be customized.
When you select document type - ZNB,
1. Internal number range is assigned.
2. Restrict item category or account assignment category.
3.If you want tax code to be flown from info record,you can make it as display mode in screen selection.(It is based on requirement of client or customer)
4. You can also fix which fields to be mandatory or required or Display.(Based on requirement)
