If you still have issues, even after the user is unlocked and password is reset in SAP, just restart your portal server(may be because of cache issue).

The user used for maintaining JCo destinations is locked or changed his/her password.
Ask your BASIS Team to reset it and set it to Password Never Expires, then edit the JCo from Portal Admin and enter the new password.

Kindly unlock the user wich you have used in the JCO connection and reset the password for the user , also update the password in the JCO connection.
