Hi , suppose I have to remove only one Transaction code like SM04 , where values given to S_TCODE is * . I have to assign this user all T CODE except SM04 . How to do it?

you have to make wildcard combination to exclude SM04
example A* to SM03* then SM05* to z* and /* elc etc
by the way removwing just tcode from S_TODE is not a very good security. You should exclude the auth object associated with SM04
