Hi ,
We have created a copy of existing layout in production and implemented some minor changes on the layout (NO ALIGNMENT CHANGES just changed one field name) .When we move the changes to testing box, the original one looks fine without any alignment issue for the output type LOCL and for the same, the new output type with major alignment issue.
One thing we what we observed is tha base version of the original layout was from (4.5B) and later it was moved to ECC. The new development(Copied one) was done in ECC by copying the original from ECC.
What are the possible causes for the alignment issue?

CHeck whether u r using the same printer in TEST and Production system .... check whether it is Letter or A4 size ect ...

If you are printing to the same device from the same system, there must be a difference in the SAPscript layout sets.
I suggest starting two SE71 sessions. Open each layout set in DISPLAY mode. Toggle back and forth between sessions and compare values.
Check the BASIC SETTINGS on the HEADER. Make sure everything is the same (page format, tab stop, font, sont size, characters per inch, etc).
Check the PAGE WINDOWS. Make sure left, upper, width, height are the same.
Check the PARAGRAPH formats. Make sure the TABS are the same. Check the LINE SPACING. Check TAB STOPS.
Another option would be to compare the output in the spool (SP01). See if they look the same when displayed.
Check the output in raw format. Select GOTO -> DISPLAY REQUEST -> SETTINGS and check RAW.
