In the log you pasted, it says....ERROR: tpsettainfo: Cofile G:\usr\sap\trans\cofiles\KMARK.STO does not exist!
You can check few points:-
1) Are the transports getting released properly and the cofiles and data files are getting generated at the OS level.
2) Try adding the transport to buffer through the OS level.
3) Check the STMS configuration again.
Thanks,
Varun

Hi ,
The system in which you are trying to add the Transport request in to the Import queue has its own Trans Directory or it is using Common Transport directory.
Can you please check with the Transport groups ?

Please check the Transport Group of that System.

Hi Gupta
Can you please check if you have a file with name <SID> in \usr\sap\trans\buffer folder in your transport domain from operating system level? There should be one file for each sap system having the same transport domain.
Open with notepad and check if entries are there..
Tell me one more thing, is it a new system you installed?
Please also tell us the systme details
Thanks and Regards
Anindya
