When it will update? During which process it will update?

J_1IINTPR2-Dispatch Register Serial number gets updated at the time of creation of excise invoice.

This is only gets updated if you have immediate utilization switched off ( I mean immediate utilization is not activated). If you are utilizing during the time of excise invoice creation then either RG23A or RG23C or PLA register gets updated according the register from which you have utilized the duty values.
