In the sales order, there have a purchase order fields on the main screen, I would like to know is it refer to purchase order? Sometimes, it is mandatory field, why need this fields, i can't imagine that create purchase order before create sales order. Thanks!

That Purchase order field is used to fill when the customer generally places the order.As it is text .
This purchase order can be taken as a reference when any misplace or any mistake happens with the company . So the customer can take the reference of the purchase order number and inform to the company
Even the company can take this purchase order number as a base and check the customers complaint.
Company can use this purchase order number field as one of the parameter to generate the user defined report

This Purchase Order field is a text field, ideally to maintain the Purchase Order Number of the Customer. In the case the order is placed verbal (without Purchase Order), it being a text field we may maintain as Verbal or probably telephonic, etc. Through configuration in Sales Order Type, the field can be be configured such that it has a unique descrition / number. In most of the cases, this field is mandatory to be maintained.

This is about the details of the Purchase Order from the customer, based on which this sales order is created. This is a maintained for reference purpose and doesn't have anything to do with the sales order processing in SAP.
