How to enable to Save Layout (Ctrl+F10)?
Is there any parameter for user, or any object to enable saving?

Well, She has enabled authorization to object S_ALV_LAYO in the user's role, although it is not recommended as per security guidelines to enable this access to endusers in the production system.
Unfortunately it won't help in my case.
My problem is specific to particular program (T-code VF44)
It seems to use a system variant. Is there a way to access and alter it?
Do you mean selection or display variant for VF44? What's the problem, don't you find the path to save a variant or are you not authorized to do it?
Within VF44 there is no special authorization to get a variant or to save a variant. The authorization is defaulted with the tcode.
