EP Administrator will manage portal content objects and applications using user administrator,system administrator ,content administrator roles to perform admin specific activities
Portal developer will develop objects using PDK,WEBDYNPRO,VISUAL COMPOSER,NWDI development tools

To make it simple, any activities within the Portal which relates to Portal administartion and Portal configuration, its the Portal Administrator. Any activities which requires building applications for the portal, thats a Portal Developer.
Which Portal elements belongs to which, I believe the best to look at is in help.sap.
