Hi Vij,
I do not think that there is any basic knowledge require for you to start Portal (checkout the prerequisites [SAP NW Portal Fundamental course|http://www.sap.com/services/education/catalog/netweaver/course.epx?context=%5b%5bEP_ADM%7cSAPEP%7c%7c%7c062%7cG%5d%5d%7c]).
My suggestion would be to take the Portal fundamental course, and then install the quick sneakpreview of the SAP Portal in your local hardware and play with it.
For learning basic Portal knowledge:
