How can we add new transportation group? After adding this new transportation group, is there any other configurations needed relating to this newly created transportation group? What are the tables related in transportation group?

The system can only carry out route determination if the transportation group is included in the material master record. You can prerequisite this by using 'Define material types'. This is the only concern after defining Transportation Groups.
One thing more you can Check which transportation groups are used in your shipping processing also.
